Hepion Pharmaceuticals stock gains focus after FDA feedback on GV104

- FDA comments reaffirmed Hepion's modeling approach while calling for revisions before the company advances its regulatory response.
- Hepion plans to address the feedback within its existing modeling program and begin commercial-scale registration batches in 2027.
- GV104 is also being developed for gambling disorder through a separate regulatory path.
Hepion Pharmaceuticals said on October 26, 2026, that written FDA feedback supported the overall approach behind its GV104 modeling program, while requesting comments on the model structure, presentation of results and fentanyl challenge simulations. The report was published by the Raleigh News & Observer.
Model results guide regulatory work
In a simulation of 200 virtual subjects, a single 16 milligram GV104 buccal film taken at least 15 minutes before exposure prevented every simulated fentanyl-induced cardiac arrest across the modeled doses. The analysis also projected a median protective onset of 10 to 15 minutes after dosing.
Those findings are model outputs rather than clinical results. Hepion said the FDA's comments could improve its ability to demonstrate long-term prophylactic protection, but the revised analysis may produce different results. The company intends to submit its response promptly.
Manufacturing remains the next step
Hepion is developing GV104 under a 505(b)(2) pathway that relies in part on prior FDA findings for an approved injectable product. The company said Chemistry, Manufacturing and Controls work and commercial-scale registration batches are the principal remaining work before a potential filing, with batch production intended to begin in 2027.
For investors, the feedback provides a regulatory development milestone but does not establish approval or clinical efficacy. The timing and outcome of any future submission remain subject to revised modeling, manufacturing work and further FDA review.
Hepion's pipeline and business
Hepion Pharmaceuticals is a U.S. biotechnology company developing therapeutic and diagnostic candidates for serious liver, immune and inflammatory diseases. Through its subsidiary Gravitas Life Sciences, it is advancing GV104, a transmucosal buccal film of nalmefene, and GV023, an oral formulation of an anti-inflammatory antibody.
The company also develops circulating tumor RNA diagnostics intended to support early detection of hepatocellular carcinoma in high-risk cirrhotic patients. Its reported lead programs address opioid exposure prevention, immunology and cancer diagnostics.
